Functional characterization of a chimeric lipase genetically engineered from human lipoprotein lipase and human hepatic lipase.

چکیده
Lipoprotein lipase (LPL) and hepatic lipase (HL) mediate the hydrolysis of triglycerides and phospholipids present in circulating lipoprotein particles and are essential for normal lipid metabolism. Both enzymes have a similar primary amino acid structure and share requirements for intact catalytic, lipid binding, and heparin binding domains. However, LPL and HL exhibit different substrate specificities and cofactor requirements. In order to characterize the functional domains necessary for LPL activity, a chimeric lipase consisting of the amino-terminal 314 amino acids of human LPL and the carboxyl-terminal 146 amino acids of human HL was synthesized by joining the cDNA of both lipases at the 5'-end of exon 7. Northern blot hybridization and Western blot analyses revealed the size of the chimera mRNA and protein to be approximately 1.5 kb and 55 kDa, respectively. The chimeric enzyme hydrolyzed both long chain and short chain fatty acid triacylglycerols and had catalytic properties that were similar to lipoprotein lipase. Thus, apolipoprotein (apo)C-II was required for maximal lipase activity, and high salt concentration abolished the ability of the chimera to hydrolyze triolein even in the presence of apoC-II. A monospecific anti-HL polyclonal antibody interacting with the C-terminal HL-derived domain of the chimeric enzyme abolished the enzyme's ability to hydrolyze triglyceride emulsion but not tributyrin substrates. Analysis of the heparin binding properties of the chimeric enzyme using heparin-Sepharose affinity chromatography revealed an elution pattern which was intermediate between that of lipoprotein and hepatic lipase. In summary, we have characterized the functional properties of an LPL-HL chimeric enzyme.(ABSTRACT TRUNCATED AT 250 WORDS)

Pseudomonas aeruginosa EF2, ATCC 9027 and ATCC 19660 were grown in a continuous culture under Tween 80 (polyoxyethylene sorbitan monooleate) limitation and optimum conditions (pH 6.5, 37°C at dilution rate of 0.05/h). Culture supernatants were carefully removed and stored at -20°C. To purify the lipases, the culture supernatant was reduced in volume to approximately 10 mL by an ultrafiltra...
متن کاملFunctional molecular mass of rat hepatic lipase in liver, adrenal gland and ovary is different.
Lipoprotein lipase (LPL) is functionally active only as a dimer. It is also generally assumed that the highly homologous hepatic lipase functions as a dimer, but no clear evidence has been presented. A hepatic lipase-like activity, also indicated as L-type lipase, is present in adrenal and ovary tissues. This enzyme is thought to originate from the liver and to be identical to hepatic lipase. W...
متن کاملMolecular Cloning and Characterization of a Lipase from an Indigenous Bacillus pumilus
Cloning and sequencing of a lipase gene from an indigenous Bacillus pumilus, strain F3, revealed an open-reading frame of 648 nucleotides predicted to encode a protein of 215 residues. Sequence analysis showed that F3 lipase contained a signal peptide composed of 34 amino acids with an H domain of 18 residues. A tat-like motif was found in the signal peptide similar to some other Bacillus pumil...
متن کاملLipoprotein lipase and hepatic triacylglycerol lipase activities in peripheral and skeletal muscle lymph.
We studied the interstitial fluid concentration of two lipid-metabolizing enzymes (lipoprotein lipase and hepatic triacylglycerol lipase) to determine their importance in interstitial modification of filtered lipoproteins. Despite the use of a very sensitive lipase assay (1 nmol of fatty acid release/ml/hr), lipase activities in plasma and in peripheral and skeletal muscle lymph from control do...
متن کاملIsolation, Optimization, and Molecular Characterization of a Lipase Producing Bacterium from Oil Contaminated Soils
Lipases have many applications in biotechnology, thanks to their ability of acylglycerides hydrolysis. They alsp possess the unique feature of acting at the lipid-water interface, which distinguishes them from esterases. Commercially useful lipases are produced by microorganisms with the extracellular lipase being produced by many bacteria including Pseudomonas. The greatest production of lipas...
